a stock solution is diluted serially by 1 in 25, then 1ml of this solution was added to 20mL of buffer solution, what is the overal dilution factor? what is the main problem with the 2nd dilution

Dilution factor 500? i.e. final solution 1/500th strength of original.
I would have thought main problem with second dilution is that you're getting a homeopathic stylee stregth to work with.
