I see. you will probably be destroying another business, if one of the multiples nearby, then go for it. I am surprised that the Drs have just given you a lease without demanding a share of the business. If, instead, they are charging you an extortionate rent, then how about a rent free three months?. At the moment looks as if you are taking all the risks and the Drs just take the money.
What about security when you are open and the surgery is closed, or will you be one of the 100 hour pharmacies where the door is locked but the pharmacy is nominally 'open'.?

If you're self employed, you can work for as long as you like. However would you really want to work 100 hours a week in a surgery based pharmacy? At one point i was doing 52 hour weeks in a busy dispensary and found it immensely tough. Do you honestly think that you will be able to run a safe and effective service on your own? Best to use either a locum or employ a manager for the hours you won't be able to physically work.
As regards wholesaler credit - it depends entirely on the wholesaler, your business model and how nice you are to them. Some of the big wholesalers have no problem supplying a 100 hour pharmacy as long as they have a reasonable business plan / income stream. Some won't touch you with a barge pole - especially if you've opened up across the road from one of their existing customers.

100 hours a week do you live next door to the surgery?

I will just re-iterate. Do you honestly think you will be able to provide a SAFE and effective service towards the end of a 100 hour week in a surgery-based, likely high volume pharmacy?
It's widely recognised that junior doctors often struggle with working 90 hour weeks. You'll be doing longer than that. Working seven days a week. How long until you get so tired you start making errors? How long until you realise that you want a life outside of the dispensary to do things like eating and sleeping?
For your own sanity, you should look into hiring another pharmacist to cover at least 40 of those hours.
