If you're self employed, you can work for as long as you like. However would you really want to work 100 hours a week in a surgery based pharmacy? At one point i was doing 52 hour weeks in a busy dispensary and found it immensely tough. Do you honestly think that you will be able to run a safe and effective service on your own? Best to use either a locum or employ a manager for the hours you won't be able to physically work.
As regards wholesaler credit - it depends entirely on the wholesaler, your business model and how nice you are to them. Some of the big wholesalers have no problem supplying a 100 hour pharmacy as long as they have a reasonable business plan / income stream. Some won't touch you with a barge pole - especially if you've opened up across the road from one of their existing customers.
