    Adjuvants in Vaccines

    Hello,

    I am not sure if this is the right place to post this, but I will still do it. I was doing an experiment combining two vaccines. One is a commercial vaccine (porcilis pesti) from Intervet that contains liquid paraffin as adjuvant and the other one is an experimental recombinant vaccine that uses 1mg Quil-A as adjuvant.
    I vaccinated pigs with both vaccines, one group received the vaccines in separate injections and the other group received the vaccines together in one injection.
    When I measured total IgG induced by the experimental vaccine in both groups I noticed that the group that received the vaccines injected together had more than a 10th fold reduction in the mean antibody titre when compared to the other group.
    Now the question is, Is there any chemical reaction or something similar that could explain this? I have been searching but I can not find anything related. Any help or idea will be much appreciated.

    is this a real-world research experiment, or is this just a re-worded university assignment?

    just a thought - quil-a is a fraction of 25-ish proteins which acts as immunologic adjuvant. liquid paraffin is a chemical adjuvant. Could there be a chemical interaction between the paraffin and some of the proteins - maybe they get denatured somehow? The processes involved in adjuvants in vaccines is very poorly understood. You might get some benefit from talking directly to one of the vaccine manufacturers - especially the formulation teams.

    Yes, it is a real world study
    I have been testing many things (like adjuvant dose, antigen concentration, storage of the vaccine, etcetera) with the experimental vaccine and one of them was the adjuvants. I had a similar idea, I was thinking that the 2 adjuvants together they may react somehow, the problem now lies on how they react. I 'll keep searching and thanks for helping me.
