Because Ambisome is made up initially from a powder and then diluted for infusion it has to be filtered to remove potential particulate matter (particles) before infusion.
This is taken from a general article on particulates in drugs and food:
"Inorganic particles in parenteral drugs are reported to include insoluble material such as glass, metal, iron rust, boiler scale, carbon, and asbestos introduced from a variety of sources.
Particles in parenteral drugs come from two basic sources. These are the drug itself, and the container and closure. Those particles intrinsic to the drug may result from contaminated make-up water, precipitates in the solution, impurities, lack of adequate filtration, migration of the filtering medium (Fig. 3), and particle entrapment and subsequent release occurring in the plumbing that handles liquid drugs." From This article (CLICK)
"Caution: Do not reconstitute with saline or add saline to the reconstituted concentration, or mix with other drugs. The use of any solution other than those recommended, or the presence of a bacteriostatic agent in the solution, may cause precipitation of AmBisome."
I would guess this is significant.
Also there may be undiluted particles of drug as well as the fact that it is a two stage dilution increasing the chance of contamination from the containers it passes through.
